The weeks are going by quickly! This week was spent finishing up the R markdown that will allow us to analyze the data we get from running our imaging videos through suite2p. All of these are new tools for me, so definitely a bit of a learning curve. We imaged the male mice from our experiment on Wednesday. They all have cranial windows and a headplate, and are imaged without anesthesia under the 2P microscope. I've used a 2P before but never for calcium imaging, so new software, and lots of new tricks learned for what to do when the microscope is acting up! Not all of the mice had good imaging due to gliosis, and some it seemed like didn't have enough expression, but some of them were excellent. Plenty of cells, bright, and watching the playback of the cells blinking was so cool. Some of them were very clear and we could even see their projections. Next steps will be to image the females next week, and see what sort of data we can obtain.
